BCG grows better with 5% CO2, but you can incubate it without on Middelbrook plates.
Since it is a slow growing mycbacteria, it will take on average 2 to 3 weeks if subcultured from an aliquot. OR 4 weeks if cultured from clinical specimens or tissus.

I used an 5% CO2 incubator for my Middelbrook plates, but in many recent works it was not reported the use of this. I started from a pharmaceutical preparation, so I hope to see a growth around three weeks. Can I ask you to report me some bibliographical reference about this?
M. bovis BCG is a very slow growing mycobacterium as rightly said and aerophilic but grow better with 5% CO2 in media. Sometimes it takes upto 6weeks for it grows, pls be patient and extend the timing in case there is no any growth seen at 3-4weeks. Tnx
