I tried to make a BCG colony count.
I used Meddlebrook agar plates and my rpincipal doubt is if i should use an incubator with or without CO2.
How long on averange BCG takes to grow?

BCG grows better with 5% CO2, but you can incubate it without on Middelbrook plates.
Since it is a slow growing mycbacteria, it will take on average 2 to 3 weeks if subcultured from an aliquot. OR 4 weeks if cultured from clinical specimens or tissus.

many thanks for your reply.
I used an 5% CO2 incubator for my Middelbrook plates, but in many recent works it was not reported the use of this. I started from a pharmaceutical preparation, so I hope to see a growth around three weeks. Can I ask you to report me some bibliographical reference about this?

For diagnostic purpose, we do not incubate Under CO2 conditions. and BCG grows well.
incubation of solid or liquid media Under 5 - 10 % CO2 results in an increase of yield and rate of growth.
