I`m trying to grow mesenchymal stem cells from umbilical cord tissue. I`ve read several protocols and i´m starting to do it myself. However, it has been over a week and i dont see cells attached to plastic and i can not tell if the technique have failed or i just have to wait longer. Have you got any experience on this?

Well we do work on isolation of stem cells from umbilical cord, but the appearance of migrating cells is not fixed. It depends on various factors:
Most likely, the appearance of migrating fibroblast like cells starts appearing in a span of 12-15 days. Initially they will appear rounded at some sides of the explant and later on will change the morphology to fibroblast like and start making colonies.
Then you can remove these explants and let the cells grow with media changes at regular intervals.Provided the probability of migrating cells is less in compariosn to the number of samples.

Thank you very much for answers! I’m trying to obtain cells from Wharton jelly. I’ve tried several protocols, explants removing vessels, explants with out removing vessels and a longitudinal cut exposing Wharton jelly with vessels to the cell plate. 13 days have passed since I did these protocols. Today I saw fibroblast-like cells for the first time as Purva Predicted! I found these cells in all techniques applied. Now I need to wait and see if these cells keep growing!
