I use TNF alpha to activate neutrophils in my experiments. I realised I've been using it too strong, however I've noticed the neutrophils always get 'sticky' after a half hour TNF alpha incubation (the most recent incubation i was deliberately trying to kill the neutrophils) but I don't see this with anything else I use (IFNs and DMSO)
Has anyone else seen this?

My former group also had a similar result. We activated human neutrophils with plant lectins. This led to incresed CD11b expression and cells stuck to cell plate just like macrophages. This became a problem for flow cytometry assays. We couldn't trypsinize cells nor use EDTA because cells started to die.

yes, I do have quite high cd11b on the neutrophils after TNF alpha incubation. I always undetstood that 'stickiness' was a result of priming/activation. Thanks for the clarification
