I transfected siRNA to the TC28a2 cell line several times. But it only has about 10% transfection efficiency. I tried the attached transfect and reverse transfection.
The experiement conditions:
6/12ul LTX
2.5ul plus reagent+siRNA
6wells plate, 200K cell
incubate with LTX mix 4hours in only DMEM media, then change to complete media without antibiotics. Check transfection efficiency after 24hours.

Srinivas Kasulla Thanks a lot for your reply! In the paper that you have mentioned, they first digested the cells with Pronase, Collagenase, and 4U/ml Hyaluronidase. I was wondering is that because they are using primary chondrocytes?
You can call Invitrogen and ask them if they ever evaluated the transfection efficiency in the cells of your interest. If they have the data they will share it with you.
Not all cell lines are Lipofectamine-friendly. So, if you tried different transfection conditions and did not get what you want, I would suggest you to switch to viral-based transduction.
