I was culturing them for the past 9 days and was about to passage them after 2-3 days. However, I observed a sudden decrease in cell number and clump formation.
I have never seen like this. Were they growing first 9 days well?

It was documented that temperature and calcium ions affect aggregation of mesenchymal stem cells in phosphate buffered saline.

Yes. They were growing and exhibiting the morphology as they should. But, all of a sudden they started to form aggregates.
