Semih, why would you do it, when you can get a deletion mutant within a week? If you want to see specific effect of knockdown of this gene, you can express this gene in the deletion strain from a plasmid driven by nmt1 promoter and you can precisely control the level of expression with thiamine. I would like to know your rationale behind such a goal.
