I would like to include a GFP tag in the plasmids I am preparing. I found multiple sequences for GFP and I would like to understand the differences between them. Does anybody have a preferred sequence?
There are a few different varieties of GFP that have been generated over the years. Have a look through the "family tree" if you're interested (https://www.fpbase.org/protein/avgfp/).
The original GFP has been almost entirely replaced by EGFP (https://www.fpbase.org/protein/egfp/) in most plasmids due to its better expression in mammalian systems. This is what I would recommend if you are simply using it as a fluorescent reporter on a plasmid.
If you are planning to create a GFP fusion protein, I would recommend using monomeric EGFP (mEGFP; https://www.fpbase.org/protein/megfp/). mEGFP does not have the weak dimerization activity of GFP/EGFP that could disrupt the function of your fusion protein.
