I am synthesizing a nematode gene for reexpression in human cells and functional characterization (not for protein expression/purification). What are the pros and cons of codon optimization in this particular context? Is it worth taking advantage of codon optimization or would it be wiser to stick with the original DNA sequence?

Hi Norbert,
Codon aptimization is only for better experssion of desired gene product. It won't effect either protein structure nor fuctional properties. I hope there are no disadvantages for the experiment. Codon optimization is the common methodology in structural biology as well.

The membrane-associated protein of my interest turned into soluble protein after codon optimization. So, yes there are cons as well.....
