I pretty much know these compunds (vitexin, isovitexin, orientin, isoorientin) might have in my sample extract. If can, that means I need to prepare four calibration curves instead? For now I am not sure to choose one of the standards or do all four for my calibration curve. I am planning to use microplate reader for estimation of these flavanoids.

Usually rutin or quercetin are used as standards for total flavonoid determination, because they are easy to find on the market and they are quite cheap. However, quantification could be performed using also other flavonoids, but they are usually more expensive than rutin or quercetin. Hence, I suggest to perform your analysis using one of these two.

Any determination of the total amount of flavonoids in terms of an individual substance is always not completely correct because the flavonoids are all different. You should decide what you want to do? if you define these connections, then of course you need to use your standards.

It is very important that rutin and quercetin are among the widespread and not unique compounds for plants. Therefore, at least you will be sure that the standard corresponds to the extract
