I am seeking guidance from investigators with experience using Percoll density gradients to isolate cells. I am trying to determine if Percoll Plus will be better for our application compared to Percoll.

I am planing to use a discontinuous Percoll gradient to isolation leukocytes from damaged muscle samples. The protocol that I'm following says to suspend the sample in 40% percoll underlain with 80% percoll and centrifuge at 400g for 30 min. Then the interphase containing leukocytes can be retrieved. I understand that Percoll Plus contains colloidal silica covalently coated with silane, while Percoll contains colloidal silica coated with polyvinylpyrrolidone.

Our downstream application at the present time is for flow cytometry but we do plan to do cell culture with cells we obtain using this method at a later date.

Is Percoll Plus worth the extra cost?
