Yes, I usually collect the old media with floating cells and centrifuge them together with the trypsinized attached cells. It gives a more precise count for the overall dead cell percentage as the floating cells are the dead cells after treatment.
Thank you Tamrin, That's exactly what i used to do; The problem is that i have to treat for 96 h, and i always got high amount of dead cells even in controls. I am trying to optimize the results ; I think it would be a concern of detachment by trypsin .
Can anybody please suggest optimal conditions for detachment of MCF7 and MDA-MB-231 cell lines (concentration of trypsin to use and time of incubation required) ? Both cells are seeded in 6 well plates.
