I am interested in sort blood stream forms (BSFs) of Trypanosoma brucei. So far I've been successful sorting the procyclic form cells of this parasite using a protocol that has been published (adapted from Archer et al 2011). When looking for information on (BSFs), some papers refer that it was not possible to do while others make no remark on the matter. The problem I've faced with the BSF cells is that I lose lots of cells during the propidium iodide staining protocol, although I do not know at which step am I exactly losing them. The end numbers are reasonably ok for normal flow cytometry analysis, but not for sorting. Has anyone experienced a similar problem, or do you suggest some adaptations to sort BSF cells due to characteristics in which they might differ from PCFs? Thank you all in advance.
