I am facing problem in re suspending of plga nano particle formed by double emulsion method . Plga nanoparticle were centrifuge at 10000 rpm at 4 degree C for 30 min but the pellet obtain was hard and insoluble in water. please suggest
I think you should redisperse pellet in the same volume of water (that you removed during centrifugation and washing process) or larger.
It may require long time to redisperse pellet again but, dissolving it in water ups and downs could dissolve it faster.

I think your pellet being to hard.You should minimise in rpm/time during centrifugation for getting soft pellet.Resuspend it with the help of water sonication for 2 min.Otherwise you can try lyphilizer.
You can add a very small amount of water (~1mL) to initiate resuspension. You can try to sonicate them for a couple minute (take care that the sonication is not destroying your NPs) and finally you should probably spent time on vortex ! Good luck
