CTC( Circulating tumour cells) are defined as tumour cells and at the time of metastasis, these specialised cells come into the circulation which can help to detect the cancer.
Kindly provide me with any protocol so that I can isolate the cells properly.

It depends on your downstream application. You might take MACS sorting and eliminate PBMCs with anti-CD45 conjugated microbeads and transfer the washed supernatant in stem cell or tumor conditioned medium.
What cancer cell type are you looking for? You could use cell sorting with anti-EpCam (which is quite specific for CTC).
Otherwise sort them out with beads against the particular and unique epitope of your CTC...
You can also use Ficoll paque to separate cell types, although the following steps depend on what your downstream application is as mentioned above - how pure you want your samples to be.
