I am treating my cells with a hypoxia selective drug, which binds to glycolytic and cytoskeletal proteins (actin, tubulin). When I trypsinize my cells after treating them with drug under hypoxia, the cells attach very poorly. This, as a consequence, gives me an amplified response. Hypoxic cells are extremely sensitive to inhibiton of glycolysis. Could that be a reason why these cells fail to adhere after trypsinization?
