When i set up my culture from a plate and do a spot test to check the presence of phages, the culture is not able to grow in the plate and I am unale to see the spots. The culture reaches log phase but after a few hours the culture disappears. What can be the possible solution to this problem?
Arnav Padhi the description of what you did is very confusing. Please provide more details so that we can better understand what you did and what happened. But if I understand that you have cultures that are not growing well or starting to grow and then disappear, if so this is usually a sign of phage contamination of the culture.
Does a single plaque form appear when performing a plaque assay? or is the spot test favorable to the same primary host bacteria as during isolation? For example, in the plaque assay and spot test using the same host bacteria as the isolation and plaque appears in the plaque assay, there is a possibility that when harvesting Phage there is still bacterial contamination so no clear spots appear. If the purpose of the spot test is to test the host range and the host bacteria are different from the primary bacteria, it is likely that the bacteria used are not in the infection range of the Phage isolates tested in the spot test.
