Hello, we recently acquired a shimadzu HPLC and started our analysis through it. What I noticed usually is the peak area of our samples are very different compared to the results got from our Agilent HPLC's. Can this be possible? All chromatographic conditions are similar. The softwares used are Chemstaion is Agilent and lab solutions in shimadzu. Thanks in advance!
Dear Shehan, Peaks in chromatography result from a wide range of variables. (too many to be explained here!) And even if you know them you may not be in a position to adjust them.(Software, Detector properties, tubular, temperatur profile, and mixing mechanism etc). Some of the differences are being used by some sales persons to argue that their product is better. Some chromatographers analyse the differences and they use this knowledge to buy equipment based on parameters and not on brands names. Regards Willi
The area under peak is not a measure for goodness for the instrument. What's important is that there is a response in the detector for your analyte, no matter what the number for area under peak is. IF you were using Agilent and you have a limit of detection say 0.5 ng/ul, then you purchased a Shimadzu and you still can achieve this 0.5 ng/ul LOD, then you are fine.
Make comparison based on the ppm, ng/ul, mg/g, or whatever unit you are using for LOD, because this is the important factor for the instrument, to be sensitive for low concentrations.
From my perspective, it would be sensible to perform a full calibration of your analyte(s) and compare the curves and their statistical evaluation.
