I need to compare data from the spot blot assay and immunofluorescence- (Integrated density from both) to determine the best method of analysing protein synthesis in serum (variable 1) and serum free (Variable 2) conditions for culturing MSCs.
What way would be best to present this and what would be the best statistical test to use?
I'm uncertain what would constitute 'best' in this case (I'll presume more signal=good). A two-way ANOVA may be ideal. In a two-way ANOVA the effect of both Serum Condition and Detection Method would be analyzed independently, and it would also evaluate the Serum-by-Detection-Method-Interaction.
If the differences are significant, you could use these data to argue that one Detection Method produces higher signal than the other, and that one Serum Condition produces higher signal than the other, and that the optimal combination of Detection Method and Serum Status produces is synergistic (e.g. it's higher than would be expected simply due to the combined effect of each independent optimization).
This is a more-reasonable method than running several T-tests, which cannot check for an interaction in any case.
There are more sophisticated means of evaluating superiority and non-inferiority, but those are beyond my understanding and abilities.
