Does anyone know if Insulin (glargine specifically) affects the OPG DUO set ELISA assay in any way? Has anyone conducted control experiements in this regard?
Dear Hannah, there are two answers for your cleaver question.
if your question related to direct interfere of glargine in OPG Duoset ELISA method, answer is no. The sandwich ELISA kit use specific antibody against Recombinant Human Osteoprotegerin/TNFRSF11B and glargine has no cross reactivity in this assay.
But if your question related to changing OPG which is measure in Duoset ELISA during using glargine in diabetic patients, the answer is yes.
Because plasma glucose has a correlation with OPG and by using glargine, FPG, HbA1c ,....affected nad these parameters change OPG (reduce OPG expression in this condition).
below paper may help you in this deduction:
Effect of pioglitazone on serum concentrations of osteoprotegerin in patients with type 2 diabetes mellitus.

Thank you Medhi for your relpy. My question was in relation to the direct interference of glargine with the assay so thank you for confirming that there is no cross reactivity with the antibody.
