I'm getting a really strange Nanodrop spectrum after Trizol extraction of RNA. Has anyone seen this kind of spectrum before, and how would you interpret it?
That big peak at 230nm is residual ethanol, or similar organic contaminating your sample. It will affect PCR/or Seq experiments. Your sample can be cleaned up either by re-precipitation or passing it through a mini prep spin column.
Hi guys, thanks for the tips. Yes, i sometimes can get rid this peak using NaAC/EtOH precipitation, though not always. The thing is, the 'cleaned' RNA can be used without problems for regular qRT-PCR, but it doesnt work so well when i do in vitro translation. Something else is retained in the otherwise clean looking RNA that inhibits IVT. Alternatively, the RNA has been modified in some way that inhibits the reaction.
