I isolate the monocytes from the peripheral blood and then I want to analyze the different phagocytosis ability in the subpopulations of monocytes by using surface antigen stain and pHrodo E.coli bioparticle. I try to do the 8-colored FACS compensation but the result is terrible. Does anyone know how to do 8-colored FACS compensation for surface antigen and pHrodo E.coli bioparticle?

Can you provide a little more information about your setup? Which fluorochromes/antibodies are you using? Which lasers and optical filters does your machine have?
I would try not to use fluorochromes that need compensation against pHrodo, which is rather easy if your setup has a yellow laser (~561nm)
