There are beliefs that iliac crest bone marrow provides superior MSCs than femur does. I collected bone marrow of the femur from patients undergoing TKR or hip surgery. Stem cells isolated from these marrows undergo chondrogenesis, adipogenesis and osteogenesis differentiation and/or exhibits markers. Any thoughts on this issue?

we analyzed human MSC from both sites a lot. There is a lot to consider:
In our first Project we compared cell from iliac crest aspirate with cells from the femur taken by a RIA (reamer/Irrigator/Aspirator). We found a higher concentration of MSC (CFU-F), early EPC and CD34+ cells in the femoral samples. Moreover,cells obtained with a RIA exerted a much higher osteogenic capacity (calcium Deposition) (Henrich et al, Injury, 2010).
Currently we adress the influence of the isolation procedure on gene expression and differentiation of the resulting MSC. We compared MSC from the femur taken either with a spoon or with a RIA and iliac crest derived MSC. Interestingly, gene Expression of osteogenic genes was highest in iliac crest MSC whereas the calcium Deposition and Deposition of elastic fibres was significantly decreased in comparison to MSC taken with the reamer. The MSC taken with a spoon were in between.
The results are not easy to discuss. Here are some of my suggestions: The RIA procedure exert a strong mechanical stress which lead to the altered osteogenic potential. The RIA procedure enriches probably other cells (partly from the inner corticalis) which have an increased osteogenic imprinting.
To summarize, it depends a lot on the procedure of Isolation. To compare equal cells one might consider to take samples with the identical procedure. If you want a high osteogenic potential and have access to a RIA then I would prefer femoral MSC.

Thanks for your e-mail and providing your valuable experience. I will think in more details about the work you have done on comparison of marrows. In my case I already did one experiment and used suction needle attached to trap containing heparin. That was used to extract marrow from patient femur (Clotting was minimal). I will think more about future experiments taking your valuable information into consideration.

I am not a pro in the field,but then i have a few agreements with Dirk! I strongly believe the procedure and the volume of marrow collected does play an important role in resulting actual MSCs which differentiate well into the 3 standard lineages.The CFU-f is the best initial way to screen.
