We are doing 10X single cell sequencing using cryopreserved cells from lymph nodes of lung cancer patients. After thaw, there is around 50% viable cells. Do we need to treat cells with DNAse to prevent the cell clumps before sorting the live cells? Thanks so much.

Treating DNAse does not hurt. But filter the whole preparation through 70um filter twice to get rid of clumps and get single cell preparation.
Also freezing and thawing will alter the gene expression. It is always good idea to do 10x on fresh cells
