Try using Papain. For me, 1 mg/ml of Papain + 1 mg/ml BSA in low Ca buffer at 37oC works the best. Incubation time has to be selected individually, it depends on many factors. Start from 10 minutes, and if tissue is not digested, increase time to 15 minutes and then to 20 minutes if necessary. In order to help endothelial cell release, I cut-open the vessels longitudinally before enzymatic digestion, and gently pipette after the digestion in fresh buffer sollution. This method gives nice endothelial sheets that are functional. I hope this is helpful!
