We want correlate results from absolut quantification qPCR to direct parasitological method (Benner,1960).
I am fairly sure the ITS-1 (not really a gene) is single copy in this single celled organism with one mitochondria.
The complete T. vivax genome sequence is available at the TriTryps database (www.tritryp.org) from where you can also do a blast search of the T.vivax ITS-1 sequence (http://www.ncbi.nlm.nih.gov/nuccore/EU482082.1) against the complete trypanosome genomes. The result will give you a single hit with the following score:
TvY486_03 | organism=Trypanosoma_vivax_Y486 | version=2013-01-1... 511 3e-143
The fact that there are no other hits with the T. vivax genome indicates that there is only one copy of this gene in T. vivax.
Hello Fabiano. In the strict concept of what is a gene, ITS1 it is not. Now, as ITS1 is a spacer sequence flanking the 3’- end of the SSU rRNA and the 5’-end of the 5.8S rRNA gene, we should expect an equivalent numbers of ITS1 copies as ribosomal copies exists. The ribosomal gene is a multicopy gene.
