I am using the "differentiated" THP1 cell line (PMA induction) to try to study IFNbeta response to PIC_LyoVec (a transfectable dsRNA mimetic). We get a significant response by qPCR for transcription.
I have not been able to show released protein by either a capture bead flow assay from biolegend or the R7D ELISA plate Duo set. I am really good with the R&D duoset on other targets.
Is there anything really useful to know about IFNbeta detection in conditioned media? For instance, I have worked with IL1beta and the transcription and release of protein are most certainly independently regulated in macrophage like cells.
Please advise.
Are you working with fresh samples? Frozen/thawed samples can be a head ache due to denaturation of proteins..
