This is NOT POWER UP that I am asking about.
Can I set up a plate Monday the 28th in the afternoon and run it Tuesday the 28th in the morning?
If yes, how should I protect or store the plate?
You seal the plate with aluminium foil to protect from light and can store at 4C overnight.
I use to make several plates and freeze at -80C.
*Seal the plate with microseal film B or whatever you are using.
*Freeze at -80C. you can use any day.
*Now take out plate from Freezer and thaw it, Spin it for 3 mins, remove the microseal film gently and replace with new microseal film and
*spin the plate one time more,
You are good to go

As I have said, it has worked for me, try it yourself before criticizing, sir. Taq polymerase are stored at -20C, anyway.
You can do it, the cDNA and the SYBR Green reagents are stable at 4C, however cover it up with aluminium foil so it doesn´t lose fluorescence. However the ideal is doing it righ before the qPCR run so you can be sure that you have the maximum quality and the most reliable results! Good luck!
I agree with @ Diogo. Store for as little time as possible. If you can, please try to finish your prep and run same day. I typically prep on ice.
