My group was the first to publish that retinoids in combination with BDNF induced differentiation in the SY5Y neuroblastoma tumor cell line in 1995. I would recommend differentiation with uM (microMolar) not mM (milleMolar) concentrations.
Helloy, there is many protocols about that. People mainly do it with Retinoic acid(RA) or RA + BDNF.
I attached an article that I am kinda following it.
"Briefly, SH-SY5Y cells were cultured with 10% complete medium (minimum
essential medium, DMEM, 10% heat inactivated fetal bovine serum, L-glutamine (2 mM), penicillin (50 U/ml), streptomycin (50 mg/ml)) in a humidified, 5% CO2, 37C incubator. Forty-eight hour after seeding, serum levels of the medium were reduced to 3%with RA (10 mM) for differentiation for seven days prior to treatment."
We also use RA, but our group just published a paper also using PMA (also called TPA), see the following link to our recent paper right here on Researchgate.
Good luck!
