Is it possible that just microscope view of seeded cells in 96 or 6 well plate can give proof (for ourselves not for paper) that the cytotoxic drug caused less cell numbers? It it can't be done as we don't count the cells? I do other assays as flow and western but just wanted to conform before proceed.
I did something similar during my undergraduate project, incubating few compounds on lymphocytes to check viability under the microscope before molecular work. I don't think it's accurate by using only your observation and microscope. That's not quantitative.
Simplest is probably using trypan blue dye exclusion test. You can start with 1 high and 1 low concentration against control, and also check if you can achieve reproducibility in the experiment. It only takes 20-30 mins to complete a n=3 by counting.
Or, try to get a simple and fast protocol (for e.g AlamarBlue and MTT) to screen your drugs' cytotoxicity. But you will need a plate reader.

For sure looking at cells under the microscope will help you determine any changes in cell confluence and morphology, but this remains qualitative as it needs to be verified using quantitative assays such as cell growth and viability ones.
