I have gone through different protocols to prepare 4% PFA for perfusion, now I am confused, which of these PFA preparation method I should follow:
a) 4% PFA in 0.1M PBS, pH 7.4 or
b) 4% PFA in 1x PBS, pH 7.4 (if it is 1X, then the molarity of PBS is 0.01M)
Will the experiment be affected, if I use 4% PFA prepared in distilled water (i.e without PBS ) pH 7.4.?

Before perfusion use heparin and sodiumnitrite to enlarge vessels. Using distilledwater induces osmotic shock and bad mrophological results. Look into Feirabend et al. J.Neurosci methods 1994, 55:137-153 and Methods 1998, 15: 123-131
