We are isolating DNA from wax embedded Squamous cell carcinoma (SCC) tissues. We take tissue sections and isolate the DNA from that. We following 3 methods for a comparison study. Those methods are Microwave method, Salting our method and by using QIAamp DNA Mini Kit from QIAGEN. My question is among the 3 methods we get lesser DNA yield in microwave method then compared to other two methods. So, what could be the reason for the lesser amount of DNA in microwave method.
