How much ethanol could be added in MS media for plant tissue culture and not harm them? Or maybe ethanol from 2,4-D solution vapors from media when added in hot one?
2,4-D is disolved in EtOH with two-three drops only, then you'll add water for final volume. You dont need to prepare it too much (max 10 mL stock with 1000 ppm is enough). Once you put it in fridge, it generaly crystalized within 2-3 days, then you can heat it up. Thats it
you could use dimethyl sulfoxide(DMS O) for dissolving 2,4-D,then complete with distilled water, or use few drops of 0.1N NaOH, then complete with distilled water.
since 2,4-D is used at very low rates , the thing to do is dissolve in as little ETOH as possible to make a concentrate , then dilute the concentrate to the final ppm of 2,4-D needed with water , what you will have formed is an aziotrope with the 2,4-D tagging along with the larger volume of water . Only a little is required and the ETOH loses most of its reactivity due to the water
we prefer to dissolve 2,4-D in few drops of 0.1N NaOH and then add distilled water to make a stock solution and then dilute with distilled water to required concentration. my students never use alcohol in tissue culture work.
