I am looking for advice. I'm currently using Chlorpromazine Hydrochloride on my U87 cells at various concentrations (2.5ug/ml, 5ug/ml and 10ug/ml), however all concentrations are causing cytotoxcity. I am taking my concentrations directly from the stock, which has DMSO in but this is below the 1%. If anyone has used this inhibitor before and could offer advice I would be very grateful.
It's been a while since I've used CPZ, but it seems as though ur lowest conc. of ~8 uM is right around the IC50 for inducing apoptosis after 24-48 hr in some cancer cell lines. So, it isn't surprising if you're seeing ~50% cell death. Is there any reason you can't dial down the conc? We typically made a 1000x dilution from DMSO. You might consider 1:1 or 1:2 serial dilutions from 10uM and measure viability at 24 h and 48 h using the MTS assay or something similar.
