Long story. I can't use any normal endogenous control in for my westerns, so I decided to go with a whole protein stain with amido black. The amido black solution I purchased has isopropanol in it and it seems to react with the membrane (0.15 amido stain, 10% acetic acid 25% isopropanol). The second I put on the stain solution it shrinks my membrane and warps it. I did a test with and without the methanol and the shrinking is due to the methanol in the transfer buffer. I also tried drying the membrane and rinsing several times in milliq. The easiest thing would be to make my own stain made up in methanol, but the other way I image would be using isopropanol to matrix match the stain solution. Thoughts?
