My reference paper says to mix ABTS solution (mine is already prepared) with a potassium persulfate (2.45 mM) solution. Can anyone provide instructions on how to prepare the potassium persulfate solution? Also, how much of the potassium persulfate solution and how much ABTS solution do I combine before incubation?
I used 7 mM ABTS soln. Weigh 0.0384 g of ABTS and dissolved in 10 mL of D H2O. The added 6.6 mg of Posassium persulfate to 10 mL of ABTS, which makes the final conc of 2.45 mM soln. Incubate in dark at 26 oC for 16 h. Hope this helps. I have attached paper that we published.

The method proposed by Brajesh is appropriate but the medium used for dissolving ABTS (water) is not suitable because the ABTS is stable in pH range of 3.5 to 6.5. For this reason we use sodium acetate (20 mM) solution adjusted at pH 4.5 by 0.1N HCl for dissolving this radical.

Brittany Kralik sorry if i'm here to ask questions not to answer you, but if you allow me Brajesh N. Vaidya please sur that mean that we put just 6.6 mg of persulfate of potassium directely to the solution of ABTS already prepared?? and the final concentration of the mexture will be 2.45mM ??

What we do is the following:
- ABTS solution: 192 mg of ABTS in distilled water to 50 mL. Store this solution in an amber flask.
- Potassium persulfate (140 mM): Mix 378.4 mg of the salt with 10 mL of distilled water. Store in the amber flask
ABTS*: The day before the experiment mix in another amber flask 5 mL of the ABTS solution with 88 uL of the potassium persulfate and leave it at room temperature for 16 hours.
Then you have to dilute 1 mL of the ABTS* to almost 100 mL of methanol. Be careful in this step because you will reach absorbances around 0.750
Then you have to add amounts of methanol to reach the 0.700 +/- 0.050. If you pass below this SD, you can add 10 uL of the ABTS* and measure again.

The answer given by Brajesh is correct. You get those values in simple calculations such as n=cV and n=m/Mw.
