I've designed several CRISPR-Cas9 binary vector constructs and I'd like to test them in tomato plants via agroinfiltration first, to confirm that they successfully cleave the genomic DNA before proceeding on to more laborious steps like tissue culture and floral dipping.
However all the protocols I've found made use of MS powder in the infiltration media recipe (5% sucrose w/v supplemented with Acetosyringone, BAP and Silwet L-77 @ pH 5.6). Is it possible to use Luria Bertani powder to make up the media instead and adjust the pH to 5.6, or is MS media the only option?
