I would like to filter AAV1 with 0.22 micron filter ,after have made iodixanol gradient and microconcentration because I saw some impurities. But I do not know if it is recommended. ( risk of decrease of titer ? or adsorption of the virus into the filter ? )
You should be fine. AAV is 20-30 nm and cellulose acetate filters are low binding. Not sure what impurities you're seeing - I'd check to make sure your production & purification are all fine. I doubt it's infectious contamination - you could always drop some into tissue culture & see if bugs grow.
Using an 0.2 um filter would be fine for contamination. For concentration, I have had success with the 4 mL or 15 mL 50 kDa Amicon Ultra centrifugal device (EMD Millipore) for concentration of 2E+07 AAV using 1500xg for 35-45 min.
