Dear colleagues I am planning to use anti-caspase 3 active in frozen brains. Have you performed that? Because I only find when tissues are paraffin-embedded. i will appreciate your answer and possibles protocols. Thanks!
You should have no problem as long as you are fixing smaller sized samples that will freeze very quickly in liquid nitrogen. Just keep everything cold until you have cyrosectioned the tissues and placed your samples on slides. Keep the slides cold until you're ready to perform the IHC. There should be very minimal to no background if you do fluorescence studies.
