I'm just curios if we can make our own cell culture media for the CHO-S cell line. I went through some of the sites for custom make a cell culture media for mammalian cells, and only Cytiva could provide a overview of the process steps for media formulations.
So my thought process was that once we understood the cell growth profile and nutrient consumption, we could custom make a cell culture media similar to commercially available media.

Basic components of a cell culture medium mainly include the following:
Monosaccharide: Cultured cells use aerobic glycolysis and anaerobic glycolysis of hexose as main energy source. In addition, hexose is used for the synthesis of some amino acid, fat and nucleic acid. Cell absorptive capacity varies among different monosaccharides, with the highest for glucose and the lowest for galactose.
Amino acid is the raw material for the cell to synthesize protein. All the cells need twelve essential amino-acids: arginine, cystine, isoleucine, leucine, lysine, methionine, phenylalanine, threonine, tryptophan, histidine, tyrosine and valine, which are L-amino acids. Furthermore, glutamine is another component playing important role in the cell metabolism process.
Inorganic ion and trace element: Besides some basic elements (including sodium, potassium, calcium, magnesium, nitrogen and phosphorus), cell growth needs some trace elements, such as molybdenum, vanadium, iron, zinc and selenium, copper, manganese.
Vitamins: Vitamins mainly act as coenzymes or prosthetic groups in cell metabolism processes. Biotin, folate, nicotinamide, pantothenic acid, pyridoxine, riboflavin, thiamine and vitamin B12 are common component in culture medium.
It would not be an easy task to prepare an in-house cell culture media as it is impossible to characterize all the variables in a biological process in cell culture. Because of their complexity, culture media can be a significant source of variability. As mentioned above, mammalian cell culture media have many components, that it may be a difficult task to finalize the concentration of each component that could provide optimum condition for cells to grow in culture.
Also, lot-to-lot inconsistency in the raw materials used can translate into variability in the performance of cell culture processes. Preparation, including the method and conditions of sterilization, may also impact media quality and adversely affect cultures.
The process of cell culture media development involves performing a sequence of screening, optimization, and confirmation. This sequence may involve a combination of methods such as automated model systems, component titration, media blending, and spent medium analysis. I would suggest that instead of designing a medium entirely from scratch, it may be easier to start by selecting a base medium from which you could work. One approach is to screen vendor media and select one with good performance. Alternatively, you may use a blend of classical media as a good starting formulation.
Once a starting medium is established, a panel of nutrient supplements can be tested. To determine the levels at which beneficial and toxic effects are seen, individual nutrients can be rapidly screened experimentally at various levels in parallel dilution series with the help of automation and high-throughput assays.

You can try preparing a simple DMEM solution, most important , FILTRE IT BEFORE USE and auroclave the solution.
