does anyone know that if Tween-20 can be used in antigen retrieval buffer in IHC experiment? And what concentration is best?
The antigen retrieval buffer I’m using now (contains 0.05% tween-20) seems show stronger staining than which contains 0.5% triton X-100 when it be used to test nuclear protein, like Ki67 but no significant difference (even a bit weaker) when to test membrane protein, like CD3. I also find it shows much more background when using buffer contains tween-20. So I want to know is there any previous study telling about the difference between tween-20 and triton in IHC experiment. Thanks!
