We would like to induce osteogenesis and adipogenesis in our hMSC lines, but in addition to traditional qualitative staining (i.e. Oil Red O, Alizarin Red, etc), we would like to analyze the degrees of differentiation by flow cytometry. Is anyone knowledgeable about how this can performed and perhaps what markers can be assessed?

Of course you can. You just need to get the markers of differentiation of these two distinct groups if cells, and bind Abs with different wavelength of light (eg. red and green). Then, all you need is to observe the amount of cells having green or red light versus the non-colored ones. That way you can separate these two distinct cells in one solution by flow cytometry and calculate the degrees of differentiation. For start, if you would be able to measure the amount of cells expected to differentiate to these two lineages, you can make an observation for their ability of differentiation,
