I would like to hear you thoughts on a very unfortunate problem that I’ve been having lately.
I’ve been doing cellular proliferation assays (MTT) to see the effect of my liposomal drugs compared to the free drug and empty control liposomes.
Unfortunately, my control liposomes are yielding an unacceptably high cellular toxicity and I cannot figure out why this is happening.
Have any of you encountered this problem before?
I've checked my cells for mycoplasma and other contaminations. During liposome production I dry the lipid film overnight in high vacuum to remove all traces of organic solvents and before treating the cells I sterile filter the lip. solution through 0.2 µm filter.
I'm working with both a DPPC:Chol:DSPE-PEG and HSPC:Chol:DSPE-PEG composition with around 5% mol DSPE-PEG in each.
I am seeing this problem when treating cells with a liposomal (phospholipid) concentration between 25 µM - 250 µM
