I am going to use Phosphate Buffer (pH 6.5) is an alternative of MES buffer (pH 6.5) for Antibody and Fluorescent Microspheres conjugation procedure. The literature I am referring to mentions MES buffer is needed to use for this reaction. Is this necessary? And what effect will PB have on the reaction?

It's probably OK, but it would help to know what conjugation chemistry you will be using.

Hi Niranjan,
If your reaction is based on EDC/NHS conjugation, there will be an advantage of using MES over PBS. Phosphate and acetate buffers may reduce the reactivity of the EDC.
