What is the difference between using phosphate buffered saline with tween 20 vs tris buffered saline with tween 20? I ask this for the application of performing a western blot as well as stripping a western blot membrane to reprobe. I usually use PBST for developing my membranes, but now wish to strip a PVDF membrane and people recommended TBST.

Actually it depends on your protein of interest. There is another thing that some antibodies like PBST, some TBST. TBS is used instead of PBS when working with phospho-specific antibodies (sometimes phosphates cause problems by interfering with anti-phosphate antibody binding to your potentially phosphorylated protein so that using PBS can result high background with these antibodies) or when working with alkaline phosphatase (PBS interferes with alkaline phosphatase also).
