We are using the neutrophil isolation kit from Miltenyi Biotec to get the neutrophils from peripheral blood from JAK2 constitutively active mice. This mouse is a leukemia model, so the traditional percoll method doesn't work. The protocol says the use of LS column is strongly recommended for bone marrow while MS column for peripheral blood. However, we only have LS columns in the lab and buying a new kit of columns and its respective separator is beyond our budget. So can I just use LS columns for peripheral blood? Will that be a big problem? Thanks everybody in advance for your time and help!
The column difference is capacity. So technically, yes, you can use the LS column. I've used both columns for the same set of beads, depending on how many cells I'm pushing through and whether it's positive or negative selection.
Bone marrow contains much more cells in general than peripheral blood (without RBCs), that's why I believe MS columns are recommended for PB, and LS for bone marrow. If you use the right for the LS column magnet, there should be not much difference what columns are used. It's more important not to overload the column with cells, and follow all the washing procedures correctly- like add new portion of wash buffer only when the previous wash buffer completely leaves the column, and never let the column to get dry. You may get a bit less cells from LS column than MS. I used both types of columns with corresponding magnets for BM, PB, spleen, lymph nodes cells population isolation without any significant differences.
