Is there a specific DNase that digest plasmid (circular) DNA and leaving linear blunt-ended DNA intact? For example after doing CRISPR-Cas9 guided digest of a large BAC vector (~100 kb), I want to separate the linear DNA from the circular undigested DNA. Some people might suggest doing pulse-field gel electrophoresis (PFGE) to resolve large linear fragments (>20 kb), but is there any other way?
I have found plasmid-safe DNase that selectively digests linear ssDNA and leave plasmid/circular DNA intact, but none for the other way round!
