I am planning a double antibody ICC on hepatocytes using polyclonal antibodies; however, one of the primary antibodies I want to use is only verified for use in IHC. Can I adapt it to be used in ICC? Would I need to lower the antibody concentration?
Yes. Definitely you can adopt the antibody for ICC. I do it all the time even if it's not validated on the product page. For the first trial do not lower the antibody concentration may be. You may optimise the concentration later if you feel like.
Many IHC antibodies will work on cells in culture or cells isolated for FACS. Some situations that would prevent ICC labeling would be things like cytosketelal changes associated with shape changes in culture, changes in secretion or receptor-mediated uptake due to loss of tissue organization, absence of a specific growth factor or cytokine in culture necessary for target protein synthesis or processing, presence of an inhibitory component in culture medium, absence of a critical extracellular matrix component involved in signaling.
