I am interested in measuring the amount of prothrombin and thrombin in the cell culture supernatant of human hepatocytes. Specifically I am interested in this kit from ABCAM.

Does anybody know if this kit or if thrombin antibodies in general can be used to detect prothrombin? Am better off using a prothrombin specific ELISA? Does anybody have any kit recommendations?
Dear Reid, I think your question is interesting. I guess you may ask the question to Abcam as, as you know, it depends on the specificity of the antibody generated. Since thrombin results from the activation of prothrombin (FII), and once activated thrombin is usually not stable (unless purified) I wonder whether you could test prothrombin by an ELISA assay (which may actually pick-up both prothrombin and thrombin antigens) and thrombin by a functional assay (a chromogenic assay or a coagulation assay measuring the conversion of fibrinogen into fibrin). It is conceivable that the thrombin functional activity in the cell culture supernatant will vary over time. I hope this helps. Kind regards, Thierry
