If the antibodies adsorbed non-specifically onto the particles, the immuno-specificity against antigen would not be accurate as high as you expect. I assume that you would like to couple them via thio-moieties at Fc region, rather than Fab, which have the antigenic sites, to lower the cross-talk. Therefore, along with the FL spectra, some immuno-assay related tools may be supportive, such as ELISA, Luminex, etc...
